Best Answer: Usually 100X (oil immersion) lens is used for stained preps. The use of oil could disturb the position of the cover slip on a wet mount. Usually 45X is used for wet mounts. If you prepare a hanging drop slide of a live bacterial culture, the oil immersion lens can be used successfully.

Are we talking about their detailed internal structures, or just to visualize their overall shape. For the former you would need an extrinsic microscope. For the latter, a standard compound light microscope or a phase contrast one would work well. For most bacteria you want magnification of about 1000x, or a 10x eyepiece and a 100x oil immersion objective lens. You also want a reasonable field of view.
Obtain it from a science catalogue. Toy shop scopes can have the magnification but are seldom good enough to be useful for this
